چکیده

This paper gives a kinetic analysis of the K(+)-selective outward-rectifier (IK,out) in the plasma membrane of Arabidopsis thaliana mesophyll cells in terms of the Hodgkin-Huxley formalism. We compared the kinetic characteristics of IK,out in Arabidopsis with IK,out channels in three other plant species that were subjected to a similar analysis: tobacco suspension cells, Vicia faba guard cells and Plantago media root cells. Because the activation kinetics of IK,out shows a clear voltage dependence, the time constant of half-activation (tau 1/2) and the elementary rate constant of channel opening (a) were calculated at the potential of half-activation (V1/2). The Arabidopsis IK,out activates relatively slowly and this is reflected in a tau 1/2 of approximately 1 s. The reason for this slow activation is twofold. Firstly, the value of a is 1.5 s-1 falls at the lower end of the range of values obtained for tobacco, Vicia and Plantago: 1.1 to 3.0 s-1. Secondly, IK,out in Arabidopsis has four closed states, while tobacco and Vicia have only two. As observed in other plant species, the activation kinetics of IK,out in Arabidopsis are sensitive to external K+: V1/2 shifts with EK but remains approximately 50 mV more positive than EK.

Existence of a delayed rectifier K+ current in the membrane of human embryonic stem cel

Introduction: Human embryonic stem cells (hESCs) are pluripotent cells that can proliferate and differentiate to many cell types. Their electrophysiological properties have not yet been chracterzed. In this study, the passive properties (such as resting membrane potential, input resistance and capacitance) and the contribution of delayed rectifier K+ channel currents to the membrane conducta...

Differences in whole-cell and single-channel ion currents across the plasma membrane of mesophyll cells from two closely related Thlaspi species.

The patch clamp technique was used to study the physiology of ion transport in mesophyll cells from two Thlaspi spp. that differ significantly in their physiology. In comparison with Thlaspi arvense, Thlaspi caerulescens (a heavy metal accumulator) can grow in, tolerate, and accumulate very high levels of certain heavy metals (primarily zinc [Zn] and cadmium) in their leaf cells. The membrane c...

Ion channels in Arabidopsis plasma membrane : transport characteristics and involvement in light-induced voltage changes.

White light (25 watts per square meter) induced an increase in plasma membrane K(+)-channel activity and a 30- to 70-millivolt transient membrane depolarization (completed in 2-3 minutes) in Arabidopsis thaliana leaf mesophyll cells. Transport characteristics of three types of ion channels in the plasma membrane were determined using inside-out patches. With 220 millimolar K(+) on the cytoplasm...

New structure and function in plant K+ channels: KCO1, an outward rectifier with a steep Ca2+ dependency.

Potassium (K+) channels mediating important physiological functions are characterized by a common pore-forming (P) domain. We report the cloning and functional analysis of the first higher plant outward rectifying K+ channel (KCO1) from Arabidopsis thaliana. KCO1 belongs to a new class of 'two-pore' K+ channels recently described in human and yeast. KCO1 has four putative transmembrane segments...

Comparison between Arabidopsis and Rice for Main Pathways of K+ and Na+ Uptake by Roots

K(+) is an essential macronutrient for plants. It is acquired by specific uptake systems located in roots. Although the concentrations of K(+) in the soil solution are widely variable, K(+) nutrition is secured by uptake systems that exhibit different affinities for K(+). Two main systems have been described for root K(+) uptake in several species: the high-affinity HAK5-like transporter and th...
